The reason is that the inside of the new container is at normal temperature, while the processed samples are at low temperatures. To preserve cryogenic samples for a long time without damaging their activity, temperature fluctuations must be avoided. If there is no liquid nitrogen in the tank and the sample is placed directly, this temperature fluctuation may cause the sample to become inactive and lose the meaning of storing liquid nitrogen samples. Therefore, liquid nitrogen must be injected first.

It is best to pre-cool first because the container is at room temperature and liquid nitrogen is at a cryogenic of -196°C. Directly injecting a large amount of liquid nitrogen has a large temperature difference, which will irritate the container material, affect the service life, and unnecessary increase in liquid nitrogen loss.

However, if the liquid nitrogen sample storage tank is in use, there is no need to consider the order, because the sample is originally in the container and has been soaked in liquid nitrogen. Taking it out and then injecting it will cause greater temperature fluctuations. And this kind of liquid nitrogen sample storage in use will ensure that the liquid nitrogen always covers the sample, and replenishing will start when it is about to be covered. In this case, the replenishing will not bring about temperature changes to the sample, so it can Inject liquid nitrogen directly without transferring the sample.
